Bcl-2 is an antiapoptotic protein that is highly upregulated in many cancers as compared to normal cells, making it an ideal target for cancer therapy. Several Bcl-2 targeting agents have been investigated over the past 30 years, but very few have exhibited any clinical significance.
Bcl-2 is a member of the Bcl-2 family of proteins that regulate apoptosis. The Bcl-2 family consists of both antiapoptotic and proapoptotic proteins. The antiapoptotic members of the Bcl-2 family include Bcl-2, Bcl-XL, MCL-1, BCLW, and BFL-1. The proapoptotic members of the family include Bax, Bak, and BH3-only proteins.
